Prédictions par lecture croisée du potentiel sensibilisant des substances chimiques : un exercice du groupe de travail FRANCOPA sur la recherche

Abstract : Predictive methods that use structural analogy are recognised alternatives to animal testing in (eco)toxicology. Among these methods, read-across predictions are particularly interesting because they can be derived by an expert on a case by case basis without using QSAR (quantitative structure activity relationship) models that require specific knowledge in molecular and statistical modelling for their interpretation. In addition, read-across predictions can be obtained by using a small number of chemicals characterised by an experimentally determined toxicological profile. Hence, this approach can be applied to the prediction of toxicological effects for which no QSAR models are available and only a few chemicals are deemed similar to the chemical of interest. These predictions are not, however, regulated by validation principles such as those that ensure the correct application and interpretation of QSAR predictions. Prompted by these observations, we (the research working group of the French FRANCOPA platform for the development, validation and dissemination of alternatives to animal testing) devised an exercise designed to evaluate the relevance of read-across approaches for predicting the skin sensitising potential of chemicals. The results show that the knowledge of mechanisms and use of empirical similarity criteria make it possible to select pertinent chemicals for which predictions based on structural analogies can be formulated.
